It's linked. If you create a subdomain, you can always reach it by using either http://subdomain.example.com or http://www.example.com/subdomain by default.
If you only want http://www.example.com/subdomain, why use a subdomain and not just create a directory called /subdomain in your public html? That way it's also reachable by using the http://www.example.com/subdomain name.
What exactly do you want to accomplish? Maybe that makes it more clear?
